Cover: Single-molecule force spectroscopy uses ligands or substrates tethered to tips of an atomic force microscope to directly probe the surface topology and to characterize conformational states of functionally unaltered cell receptors in intact cells. In this study, direct evidence was obtained that a stereo-selective sugar-binding site appears on the surface of the Na+-glucose co-transporter SGLT1 in the presence of Na+, and that a specific site interacting with the inhibitor phlorizin is associated with conformational changes.
