Movie S1. Miranda localizes to the basal cortex via the cytoplasm during symmetric division of NE cells. Embryos expressing UAS-Miranda-GFP under the control of neuralized-Gal4 were monitored by time-lapse microscopy. Pictures from a single confocal plane were taken at 13.5-second intervals.
Movie S2. PON moves primarily via the cortex to a basal crescent in symmetrically dividing NE cells. Movie of a NE cell expressing PON-GFP, sca-Gal4 (Roegiers et al., 2001). Settings were the same as for Movie S1. As reported previously, PON moves along the cortex to the basal pole in embryonic NE cells (Lu et al., 1999).
Movie S3. Miranda shows cytoplasmic localization prior to formation of a basal crescent during asymmetric NB mitosis. In asymmetrically dividing NB expressing Miranda-GFP forms a ‘cytoplasmic cloud’ in metaphase, which moved rapidly from the apical to the basal side of the cell. The white circle indicates the larger neuroblast. Settings were the same as for Movie S1.
Movie S4. PON moves via the cortex to the basal cortex in asymmetrically dividing NBs. A movie of embryos expressing PON-GFP shows that distinct from Miranda (S1/S3), the adaptor protein PON moved primarily along the cortex to a basal crescent during asymmetric neuroblast mitosis. The white circle indicates the larger neuroblast.
