Subsequently, stable cell lines were selected and analysed for occluding expression. Chicken occludin (Huber et al., 2000) was ectopically expressed in GPNT cells and stable cell lines derived. Fig. S2. Enlarged section of Fig. 6D. To illustrate differential phosphorylation, phosphopeptides have been subdivided into two groups annotated by differently colored circles. In VEC from ICAM-1 crosslinked cells (XL) peptides from each group display reduced mobility, suggesting additional or altered phosphorylation. In Y645F-VEC, Y731F-VEC and Y733F-VEC, but not Y685F-VEC, these crossslinking-specific phosphopeptides were absent, suggesting that these mutations interfered with ICAM-induced phosporylation. Note, Y645F appeared to induce additional or compensatory phosphorylation in the peptide group marked by a blue circle.
