Fig. S2. The effects of ADMA and SDMA on cell adhesion. PAECs were left untreated (control) or pre-incubated with 100 μM ADMA or 100 μM SDMA for 24 hours prior to the experiment. The cells were trypsinized, centrifuged and re-suspended in pre-warmed, serum-containing culture medium with no inhibitors, or medium containing ADMA or SDMA. The cells were then plated at the density of 104 cells/ml in 3 cm Petri dishes covered with fibronectin. Following 15 minutes incubation at 37°C, the dishes were washed once in PBS to remove non-adherent cells and the remaining cells were fixed in a 4% solution of formaldehyde in PBS. The numbers of adherent cells were counted under the phase contrast microscope. The graph shows changes in the average number of cells in 1 mm2. The number of dishes per treatment was nine, analysed in three separate experiments. *P<0.05, comparison with untreated controls.
