Expression levels of p65dsRed and IkBaEGFP affect protein kinetics in response to TNFa. Video time series images of IkBa-EGFP and p65-dsRed fluorescence over a period of 70 minutes after addition of 10 ng/ml TNFa. Green and red fluorescence was recorded as separate images and then merged for visualisation. Green IkBa-EGFP and red p65-dsRed co-localisation is represented as yellow in the presence of a higher IkBa-EGFP:p65-dsRed ratios, and orange in the presence of higher p65-dsRed:IkBa-EGFP ratios. Those cells with a lower ratio of IkBa-EGFP:p65dsRed show faster nuclear translocation of p65-dsRed and IkBa-EGFP degradation than those with higher IkBa-EGFP:p65-dsRed ratio. Also, p65-dsRed nuclear export can be seen towards the end of the time series in those cells with a higher p65-dsRed:IkBa-EGFP ratio. The movie comprises stills taken every two minutes encoded with Sorenson video at 15 frames per second compressed to produce a file smaller than 1 Mb.
