Osmichrome Fixation

Fixation by the combined action of osmium tetroxide dichromate involves the use of two reagents which produce their best results at two different hydrogen ion concentrations. By applying them in sequence the conditions may be adjusted independently for the two reagents. For tissues of the rat the following schedule has provided good results: (1) Fix for 5 to 7 hours at 5° C in 1% osmium tetroxide buffered at pH 7.2; (2) rinse briefly; (3) chromate for 18 hours at 20° C in 2.5% potasium dichromate dissolved in 5% mercuric chloride; (4) embed in parafin.
